If you cannot manually lock the door then the actuator in the door might have seized? I had one in a previous 5, water got into it. Another possibility is that the actuator is not receiving the signal to open or close the lock. The actuators can be quite strong and it takes a lot of effort to force it to move. Check all of the connections from the alarm/central locking to the actuator.
