If you cannot manually lock the door then the actuator in the door might have seized? I had one in a previous 5, water got into it. Another possibility is that the actuator is not receiving the signal to open or close the lock. The actuators can be quite strong and it takes a lot of effort to force it to move. Check all of the connections from the alarm/central locking to the actuator.

You won't really know until you remove the door trim and have a look.

If you can still hear the actuator working, then the clamp holding the actuator rod to the door lock rod might have come loose. Be careful when you tighten the screws back as the clamp is made of soft metal. Applying a drop of Loctite 243(blue) to the screws will solve the clamping problem for good.

If the actuator is dead due to water damage or whatever while the electrical connection is ok, get a new one from Jaycar or Autobarn for about $20. While you have the door panel apart, wrap the actuator with a plastic sheet to protect it from water damage.
